In the example shown i the diagram above the 454 workflow is used. If the concentration of the sample is overestimated the majority of the beads will be blanked resulting in a failed sequencing read. Conversely, if sample concentration is underestimated, the majority of the beads will contain multiple copies of DNA which will in turn cause a failed read. Sequencing throughput and data quality is maximized when the maximum number of emPCR beads contain exactly one DNA molecule.
