The BioMark Dynamic Array - Genotyping

Genetic research usually begins with whole genome screens of a few samples with hybridization arrays to develop candidate gene profiles. But, that’s often where the progress stops. Many laboratories lack the resources to take the followup step—validation of the 30 – 300 genes uncovered in screening against a sufficiently large population. SNP genotyping by PCR is the most rigorous approach to validation; however, performed in microwell plates, genotyping is entirely too arduous in terms of material- and liquid-handling complexity. This reality leaves many good hypotheses waiting in the wings.

The BioMark 48.48 Dynamic Array is a highly efficient solution for mid-multiplex genotyping (projects that involve between 30 – 300 genes) using TaqMan® PCR assays. The key to this efficiency is the matrix of channels, chambers, and valves that do the work of assembling assays. Forty-eight samples and 48 assays are loaded into the inlets of the chip’s input frame and pressure loaded into 2,304 reaction chambers. That’s a total of 96 steps to test 48 genes against 48 samples in a single run.

The savings in time and effort add up rapidly. For example, a 2,000-sample study could be completed with only 4,032 pipetting steps. That’s just two percent of the 192,000 steps that would be required to complete the same study with microwell plates. With dynamic arrays, genetic researchers can advance their research much more easily and affordably, and they can do so while using gold-standard TaqMan® assays.
