Fluidigm Dynamic Arrays for Gene Expression

Bringing the density of microarrays to the world of qPCR.

	Global Chip View: Cts of a Real-Time PCR Run Each 96.96 Dynamic Array produces 9,216 real-time PCR results. Fluidigm's data analysis provides a global view of an entire chip's Cts so that overall experimental success can be quickly assessed. This view is color-coded for Ct ranges; black positions indicate the target gene was not detected. In this example, most of the black positions are negative controls.

	Intra- and Interchip Reproducibility Twenty-four TaqMan® assays were run against cDNA that was commercially obtained. Each assay was repeated six times on each 48.48 Dynamic Array and the experiment was repeated on three different 48.48 Dynamic Arrays.The mean Cts and standard deviations for each assay are shown in this table.

	Assay Dynamic Range on the BioMark System The ten-fold dilution series of a DNA template was assayed against a TaqMan® assay specific for the template. This graph shows that 48.48 Dynamic Arrays, when run in the BioMark system, show linearity over at least six orders of magnitude.

	Correlation Scatterplot Between BioMark System and Applied Biosystems 7900 A commercially obtained cDNA sample was run against 31 TaqMan® assays in triplicate on both the Applied Biosystems 7900 (10 microliter reactions in a 384-well plate) and the 48.48 Dynamic Array. This scatterplot compares the mean Cts of each system. The correlation coefficient was 0.99.
